Could somebody help me on making sprites from scratch?

I really need some help,I know how to shade my sprites and all that,but I don;t know how to MAKE one,not just paint one up,with ACTUAL PIXELS. If you can help me with this,or just give me some tips,that would be really helpful! (P.S. I use GIMP 2,and Im about to get photoshop)
