Making a guitar hero game

I'm trying to make a compter game for my programming class, and I'm looking for help on how to program it. I'm looking for a way to make you have to hit a button when the note hits a line, and if you don't hit the button, the music stops and you lose a "point" and the music doesn't restart until you get a note right.

Any suggestions?
