The best advice I can give you is simply this: do it step-by-step. The first thing I would do make your character, and get him/her moving around (assuming it isn't a point/click game).
Then develop their powers/attacks/abilities. Make variables for attributes like "strength", "accuracy" ,etc. Making one enemy to be your test subject can help. After you get one successful enemy, copy it and modify to make additional enemies.

But I make platformers step-by-step too like this:
A) Make a simple arena with one wall and one floor
B) Make your character interact correctly with the wall and floor
C) Polish up movement/walls. Bad movement right now will lead to glitches like moving through walls or getting stuck in them
D) Elaborate off this basic structure, make levels

so you need a guide from making an rpg with nothing right now, right? or do you have something already made?
I can guide you through it, if you want, but unless you just want tips or just want to know how to do a specific thing.
btw, your rpg will not be very long, because scratch's 10MB limit. doubt you'll get there, but just a heads up.

Aha, that I can help you with.
To make things disappear and reappear, simply use the "hide" and "show" blocks.
To change your health level, simply use a variable - if you get hit or whatever, just lower the variable, and if you get medicine, raise the variable.
Better controls are purely a result of experimentation. Just put a few blocks together and test out the controls. If you think there's something that should be changed, change it and go back again. It's pretty simple, actually.

To be honest, there are many ways to make an RPG. Some may be harder than others. But in general, they are all hard.
Making an RPG requires movement, battle system, experience, monsters, quests, and many other things, and all can be done many ways as well. Now, first you need to make a plan on how it's going to run, will it be a top-down view like The Legend of Zelda or a side view like Maple Story ( or whatever those games are
)? You'll need to make character movement, collisions with walls, or other things you don't want people to get through. Then you'll need to do the same for monsters, then allow battling, then everything else is up to you. You get to make your quests, battle moves/attacks, weapons, abilities, places, buildings, etc. It isn't an easy task, so if you get stuck somewhere, wait a bit, do some searching on how to do it, then go work on it again.
