Unfortunately, the PicoBoard does not work with projects online. You must download your project for it to work.
I've taken a long hiatus, but I still visit sometimes. Give me some time to answer any messages you post on my projects!

Flash (what Scratch 2.0 will be written in) supports external computer devices such as microphones and webcams online, so online Picoboard and WeDo support is a possibility. I'm not sure if the Scratch team is planning on adding that functionality, though, as a pretty small percentage of Scratch users have either of those.
