If you mean on the same computer, you can just add an extra sprite, and give it different controls (eg - W,A,S,D instead of the arrow keys)
If you want to make a game that you can play together with someone on another computer, take a look at Jonathanpb's guide.
The second method can get a little fiddly sometimes, though
