Hardest: Excise Psi (Not a boss, but who cares, its so hard!)
Best: Not sure. I liked most of them.
Most Anoying: Whats-His-Name and the undead dragon. Its really the Undead Dragon. It can regenerate all its health when it is dead!

sonic generations final boss. it is just flying around getting rings. i don't have the game, i just know this.

Hardest boss fight is Norman in Residant Evil Revelations. On the hardest mode.

Now, I think that one of the hardest and best bosses I've seen is Gideon from Scott Pilgrim Vs The World: The Game. There is spoilers, so highlight the text if you want to see.
You start the battle going down an elevator. You have a time limit. You can fall off. You beat up enemies, and when you stop the elevator is held down and you need to break them as enemies board. After you get down, you start the second phase, and fight Gideon. Then you get trapped in some sort of creepy mind world. You pick up a heart sword and fight the second incarnation of Gideon, which is a god-like version of himself. He is very hard to defeat in this form. Then you escape this area, and go through the final level, the techno base and the third stage of the battle. After you complete the level, Gideon falls to his knees, and the BREAK!!! sign appears near him. You destroy him and you finally see the satisfying KO sign.
Worst, is probably waddle dee. He sucks.

I saw a video of portal 2 and it's a part where Wheatley tries to make you die, the player jumped down off a cliff after a while, and he got it right the next time. Don't really remember the video. Oh, Wheatley, you're a jerk. Portal 2 is amazing. Never played it, but one guy does a lot of custom portal 2 maps and those are so funny. I liked the one where it ended in the player being dismantled and killed, even though they're not a robot. And one time, the custom map starts off with GlaDOS yelling surprise (slightly unenthusiastically) and there was maybe a little confetti, and then GlaDOS says Chell's parents abandoned her at birth, trying to make the boring surprise better (or maybe worse).
Oh man, my brother loves watching that guy playing Portal 2 custom maps.

I don't know if is the best or the worst, but the most difficult is Otane, from Power Instinct 2 (arcade fighting game). All of the fighters in this game are strong, but Otane is the stronger. And quicker. And one of her special moves (NOT a Super Move) can KO you in a shot, and lower your HP a lot if you are guarded. This is a very strong granny. I never won vs. her.

Also is Sissy, of Power Instinct Matrimelee (I like this saga). It's the less expected boss (a littyle princess), with the music less expected (the Wedding March-techno), very agile and strong. Although, is a boss Megaman-styled: she has an attack pattern, and his deffense has a lot of holes.

Ending / last level of Super Hexagon.

That last level is the hardestestestest (sic) of all of them. And the level after that is creepy, and the ending made me cry.
