topic name says it all, I want to know how to do that.

These work for Windows, but I don't know about Mac/Linux/etc.
Icon:
1. Go to your "My Stuff" page, and select "change picture". Open up "choose file".
2. Select "Computer", then the hard drive. After that, open up "Program Files", and find the "Scratch" folder.
3. Go to "media", and choose a sprite from one of the folders in "Costumes"
4. Click "OK", and it will change your image for you.
Signature:
1. Open up an image uploading site, such as tinypic or imageshack.
2. Click the "upload images" button (the words may be slightly different) on that website, and to find a sprite, follow steps 2 and 3 of the icon instructions.
3. Click "OK", and upload it. It might ask you to fill in a "Captcha", which is looking at an image, and typing the words from it. Then, copy the BBCode version of the code that comes up when it's done uploading (it should have [img] tags, with square brackets).
4. Paste it into your forum sig box (on the forums, go to "Profile", then "Personality") Click "submit".

You can't use actual scratch sprite files.

You can't use actual scratch sprite files.
so how do I covert it to make it a wroking moving icon?

You can't, unfortunately. The only option I can think of is taking a video of the screen, cropping it, and then converting that to a GIF.

Great idea, but I'm afraid uploading sprites won't work - they're not image files and can't be interpreted by anything except Scratch itself.

yeah, doesn't work, yet, I just found out that sprites are .sprite files, I just need to find a site where I can upload a .sprite file and it should work
which there are no .sprite file sites

But browsers can't display .sprite files as they aren't images. I'm afraid you can't put a sprite in your signature.

You can make an animated GIF file by using flash.
First download the trial version of Flash CS5 if you don't already have it. (I'm using Flash 8 so it might be different.)
Save every separate costume you want to use into picture files (jpg, jpeg, png, bmp, etc) and then import them into flash using file > import > import to stage.
You might not know how to do this, make the animation using the costumes among keyframes. It's basic animating in flash.
Press ctrl + enter to test it.
If it turns out OK then use file > export > export movie. Make sure you're saving it as an animated gif file.
It should be where you saved it to. You probably weren't able to do this so sorry if you couldn't; that's the only way I can make an animated gif besides gif ninja. Try that if this didn't help. http://www.gifninja.com/CreateAGif.aspx
