The "loudness" block won't do anything if you don't have a microphone connected to your computer. It senses how loud the noise going into the microphone is.
Try using the "change volume by _" and "set volume to _" blocks if you want to change the volume of an instrument instead.
I'm taking a break from Scratch until 2.0 comes out. Any messages sent between then and now probably won't be read - sorry.

Also, the loudness block doesn't change the volume - it measures it.
